In an effort to develop inducible switches allowing for flexible fine-tuning of gene expression levels in insect cells, we have compared the induction capacities of two Drosophila minimal promoters when linked to four consecutive ecdysone response elements. These minimal promoters, either containing a TATA-box or a downstream promoter element, drove the expression of a luciferase reporter gene. Potent induction capacities were observed with the insect moulting hormone, 20-hydroxyecdysone, and with ponasterone A, a plant ecdysteroid. The developed inducible switches further expand the repertoire of molecular tools for functional expression of proteins of interest in insect cells. In addition, the combination of an ecdysone switch with promoters that possess different structural elements can provide novel insights into ecdysteroid-induced transcription in an insect cell line.","is_dataset_classified":null,"base_score":2.0794415416798357,"endowment":2.0794415416798357,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"15110866","pmcid":null,"openalex_id":"https://openalex.org/W1989155943","authors":[],"funders":[],"total_grants":0,"fwci":0.304,"citation_percentile":0.4401615,"influential_citations":0,"citation_trend":[{"year":2017,"count":1},{"year":2026,"count":1}],"oa_status":"closed","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0965174804000220?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0965174804000220?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ibmb.2004.02.005","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/15110866","host_type":"repository"},{"url":"http://gateway.newisiknowledge.com/gateway/Gateway.cgi?GWVersion=2&SrcAuth=CCC&SrcApp=PRODUCT_NAME&SrcURL=WOS_RETURN_URL&CKEY=POEL0451040034IJ&DestLinkType=FullRecord&DestApp=CCC&SrcDesc=RETURN_ALT_TEXT&SrcAppSID=APP_SID","host_type":"repository"}],"fields_of_study":["Neurobiology and Insect Physiology Research","Insect and Arachnid Ecology and Behavior","Insect Utilization and Effects","Animals","Base Sequence","Cell Line","DNA-Binding Proteins","Dose-Response Relationship, Drug","Drosophila","Drosophila Proteins","Ecdysterone","Gene Expression Regulation","Genes, Reporter","HSP70 Heat-Shock Proteins","Insect Proteins","Luciferases","Molecular Sequence Data","Promoter Regions, Genetic","Receptors, Steroid","Transcription Factors","Transfection"],"mesh_terms":["Animals","Base Sequence","Cell Line","DNA-Binding Proteins","Dose-Response Relationship, Drug","Drosophila","Ecdysterone","Gene Expression Regulation","Luciferases","Molecular Sequence Data","Promoter Regions, Genetic","Receptors, Steroid","Transcription Factors","Transfection","Genes, Reporter","HSP70 Heat-Shock Proteins","Insect Proteins","Drosophila Proteins"],"keywords":["Ecdysteroid","Ecdysone","Biology","Schneider 2 cells","Promoter","Ecdysone receptor","Reporter gene","Gene","Transcription factor","Gene expression","Cell biology","Transcription (linguistics)","Response element","Genetics","RNA","RNA interference","Hormone","Biochemistry","Nuclear receptor"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-16T19:58:40.834868Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
