These PTMs play pivotal roles throughout the viral infection cycle by governing the structure, function, and host-cell interactions of viral proteins. While traditional PTMs, such as phosphorylation of the polymerase complex, ubiquitination of viral scaffolds, and glycosylation of surface antigens, remain central, recent multi-omics advances have unveiled a broader landscape, including SUMOylation, acetylation, and carboxylation. Furthermore, these modifications do not function in isolation; they form a multilayered crosstalk network that orchestrates the delicate balance between viral fitness and host immune evasion. These advancements provide crucial foundations for understanding influenza pathogenesis and informing the development of novel antiviral strategies. This review provides a comprehensive overview of the functional roles of both host- and virus-mediated PTMs during the influenza virus infection process. In addition, we highlight current research gaps and suggest future directions for targeting PTMs in therapeutic design.
