Melanosomes in the retinal pigment epithelium (RPE) are formed during embryogenesis and mature during early postnatal life while lipofuscin and melanolipofuscin granules accumulate as a function of age. The difficulty in studying the formation and consequences of melanosomes and lipofuscin granules in RPE cell culture is compounded by the fact that these pigment granules do not normally occur in established RPE cell lines and pigment granules are rapidly lost in adult human primary culture. This review will consider options available for overcoming these limitations and permitting the study of melanosomes and lipofuscin in cell culture and will briefly evaluate the advantages and disadvantages of the different protocols.","is_dataset_classified":null,"base_score":4.532599493153256,"endowment":4.532599493153256,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"25152361","pmcid":"PMC4143628","openalex_id":"https://openalex.org/W2087386497","authors":[],"funders":[{"funder_name":"National Institutes of Health","grant_id":"EY019688","title":null},{"funder_name":"National Institutes of Health","grant_id":"EY021626","title":null},{"funder_name":"NEI NIH HHS","grant_id":"R01 EY019688","title":null},{"funder_name":"NEI NIH HHS","grant_id":"R21 EY021626","title":null},{"funder_name":"National Institutes of Health","grant_id":"7R21EY021626-03","title":"Circadian-dependent autophagy in retinal maintenance and diabetes"},{"funder_name":"National Institutes of Health","grant_id":"1R01EY019688-01","title":"Autophagy: A critical factor in RPE aging and AMD"},{"funder_name":"Foundation Fighting Blindness","grant_id":"","title":null}],"total_grants":7,"fwci":3.0953,"citation_percentile":0.91594296,"influential_citations":0,"citation_trend":[{"year":2014,"count":1},{"year":2015,"count":7},{"year":2016,"count":3},{"year":2017,"count":11},{"year":2018,"count":11},{"year":2019,"count":6},{"year":2020,"count":7},{"year":2021,"count":4},{"year":2022,"count":9},{"year":2023,"count":15},{"year":2024,"count":12},{"year":2025,"count":3},{"year":2026,"count":3}],"oa_status":"hybrid","license":"cc-by-nc-nd","oa_locations":[{"url":"https://doi.org/10.1016/j.exer.2014.01.016","host_type":"journal"},{"url":"https://doi.org/10.1016/j.exer.2014.01.016","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014483514000311?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014483514000311?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/25152361","host_type":"repository"},{"url":"https://hdl.handle.net/1805/8155","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/4143628","host_type":"repository"},{"url":"http://hdl.handle.net/1805/8155","host_type":"repository"},{"url":"http://dx.doi.org/10.1016/j.exer.2014.01.016","host_type":""},{"url":"https://dx.doi.org/10.1016/j.exer.2014.01.016","host_type":""},{"url":"https://doi.org/https://doi.org/10.1016/j.exer.2014.01.016","host_type":""}],"fields_of_study":["melanin and skin pigmentation","Retinal Development and Disorders","Photoreceptor and optogenetics research","0301 basic medicine","03 medical and health sciences","0302 clinical medicine","Animals","Cells, Cultured","Epithelial Cells","Humans","Lipofuscin","Melanins","Models, Animal","Models, Biological","Retinal Pigment Epithelium"],"mesh_terms":["Animals","Cells, Cultured","Epithelial Cells","Humans","Lipofuscin","Melanins","Models, Biological","Models, Animal","Retinal Pigment Epithelium"],"keywords":["Lipofuscin","Melanosome","Retinal pigment epithelium","Cell biology","Melanin","Cell culture","Retinal","Retina","Biology","Pigment","Cell","Chemistry","Neuroscience","Biochemistry","Genetics","Rpe","A2e","Melanosomes","Cells","Models, Biological","Cellular and Molecular Neuroscience","Animals","Humans","Cells, Cultured","Melanins","Cultured","Epithelial Cells","Sensory Systems","Ophthalmology","physiology","Models, Animal","cytology"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
