Ginsenoside Rb3 (Rb3) is known to exhibit protective effects against myocardial ischemia, its impact on CIRI remains unclear. Therefore, we investigated the protective effects of Rb3 on CIRI and its underlying mechanisms. Our results showed that Rb3 reduced cerebral infarct volume, decreased blood-brain barrier (BBB) permeability, and improved neurological deficits in CIRI rats. Rb3 also mitigated cerebral ferroptosis and alleviated neuroinflammation, as evidenced by decreased iron levels, reduced MDA content, an improved GSH/GSSG ratio, and lower levels of TNF-α, IL-1β, and IL-6, through modulation of the NLRP3/NF-κB/GPX4 pathway. Additionally, Rb3 alleviated intestinal inflammation, improved the intestinal barrier, and corrected gut microbiota dysbiosis and reduced the microbial metabolites TMAO and LPS in CIRI rats. It is noteworthy that in pseudo germ-free rats with CIRI, fecal microbiota transplants (FMT) from Rb3-treated rats conferred similar protective effects as Rb3. Summarily, this study reveals that Rb3 reduces neuroinflammation and ferroptosis in the brains of middle cerebral artery occlusion/reperfusion (MCAO/R) rats via the NLRP3/NF-κB/GPX4 pathway in a gut microbiota-dependent manner.","is_dataset_classified":null,"base_score":1.9459101490553132,"endowment":1.9459101490553132,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"40876612","pmcid":null,"openalex_id":"https://openalex.org/W4413676823","authors":[],"funders":[{"funder_name":"National Natural Science Foundation of China","grant_id":"U1812403-5-3","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"U20A20405","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"81891012","title":null},{"funder_name":"Chinese Academy of Medical Sciences Initiative for Innovative Medicine","grant_id":"2022-I2M-2-002","title":null},{"funder_name":"Chinese Academy of Medical Sciences Initiative for Innovative Medicine","grant_id":"2021-I2M-1\\u2013031","title":null},{"funder_name":"Chinese Academy of Medical Sciences Initiative for Innovative Medicine","grant_id":"2022-I2M-1\\u2013018","title":null}],"total_grants":6,"fwci":2.7264,"citation_percentile":0.91326223,"influential_citations":0,"citation_trend":[{"year":2025,"count":1},{"year":2026,"count":5}],"oa_status":"closed","license":"https://doi.org/10.15223/policy-004","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0014299925008453?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014299925008453?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ejphar.2025.178091","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/40876612","host_type":"repository"}],"fields_of_study":["Cancer-related molecular mechanisms research","Gut microbiota and health","MicroRNA in disease regulation"],"mesh_terms":["Gastrointestinal Microbiome","NLR Family, Pyrin Domain-Containing 3 Protein","Ferroptosis","Neuroinflammatory Diseases","Animals","Blood-Brain Barrier","Brain Ischemia","Disease Models, Animal","Male","Signal Transduction","Reperfusion Injury","NF-kappa B","Rats, Sprague-Dawley","Ginsenosides","Rats"],"keywords":["Ginsenoside Rg1","Inflammation","Ischemia","Reperfusion injury","NF-κB","Pharmacology","Medicine","Ischemic injury","HMGB1","Ginsenoside","Immunology","Pathology","Internal medicine","neuroinflammation","Ferroptosis","Ginsenoside Rb3","Cerebral Ischemia/reperfusion Injury","Microbiota-gut-brain Axis","Nlrp3/nf-κb/gpx4 Pathway"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T00:02:00.135906Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
