In spite of its central role in physiological homeostasis, it provides the oxygen and nutrition needed by tumor cells to proceed from dormancy if pro-angiogenic factors tip the balance in favor of tumor angiogenesis. Among pro-angiogenic factors, vascular endothelial growth factor (VEGF) is a prominent target in therapeutic methods due to its strategic involvement in the formation of anomalous tumor vasculature. In addition, VEGF exhibits immune-regulatory properties which suppress immune cell antitumor activity. VEGF signaling through its receptors is an integral part of tumoral angiogenic approaches. A wide variety of medicines have been designed to target the ligands and receptors of this pro-angiogenic superfamily. Herein, we summarize the direct and indirect molecular mechanisms of VEGF to demonstrate its versatile role in the context of cancer angiogenesis and current transformative VEGF-targeted strategies interfering with tumor growth.","is_dataset_classified":null,"base_score":5.846438775057724,"endowment":5.846438775057724,"datacite_reuse_total":2,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"36906141","pmcid":null,"openalex_id":"https://openalex.org/W4323660090","authors":[],"funders":[],"total_grants":0,"fwci":31.9029,"citation_percentile":0.99889344,"influential_citations":0,"citation_trend":[{"year":2022,"count":1},{"year":2023,"count":16},{"year":2024,"count":73},{"year":2025,"count":180},{"year":2026,"count":75}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://doi.org/10.1016/j.ejphar.2023.175586","host_type":"journal"},{"url":"https://doi.org/10.1016/j.ejphar.2023.175586","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/36906141","host_type":"repository"},{"url":"https://figshare.com/articles/journal_contribution/The_role_of_VEGF_in_cancer-induced_angiogenesis_and_research_progress_of_drugs_targeting_VEGF/28678577","host_type":"repository"}],"fields_of_study":["Angiogenesis and VEGF in Cancer","Cancer Mechanisms and Therapy","Cancer, Hypoxia, and Metabolism"],"mesh_terms":["Humans","Neoplasms","Neovascularization, Pathologic","Angiogenesis Inhibitors","Vascular Endothelial Growth Factors","Vascular Endothelial Growth Factor A"],"keywords":["Angiogenesis","Cancer research","Context (archaeology)","Vascular endothelial growth factor","Immune system","Tumor microenvironment","Receptor","Cancer","HIF1A","Biology","Immunology","Cell biology","Medicine","VEGF receptors","Internal medicine","Biochemistry","Anti-vegf","Anti-angiogenic Agents","Human Solid Cancers"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Zero hunger"}],"linked_datasets":[{"doi":"10.6084/m9.figshare.26610165.v1","title":"Additional file 1 of Synergistic efficacy of simultaneous anti-TGF-β/VEGF bispecific antibody and PD-1 blockade in cancer therapy","publisher":"figshare","resource_type":"JournalArticle"},{"doi":"10.6084/m9.figshare.26610165","title":"Additional file 1 of Synergistic efficacy of simultaneous anti-TGF-β/VEGF bispecific antibody and PD-1 blockade in cancer therapy","publisher":"figshare","resource_type":"JournalArticle"}],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-04T03:37:33.342626Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
