As such, antagonism of glucagon receptor signalling represents an exciting approach for treating diabetes. To harness these beneficial metabolic effects, two novel glucagon analogues, desHis¹Glu⁹-glucagon-[mPEG] and desHis¹Glu⁹(Lys³⁰PAL)-glucagon, has been evaluated for potential glucagon receptor antagonistic properties. Both novel peptides were completely resistant to enzymatic breakdown and significantly (P<0.05 to P<0.001) inhibited glucagon-mediated elevations of cAMP production in glucagon receptor transfected cells. Similarly, desHis¹Glu⁹-glucagon-[mPEG] and desHis¹Glu⁹(Lys³⁰PAL)-glucagon effectively antagonised glucagon-induced increases of insulin secretion from BRIN BD11 cells. When administered acutely to normal, high fat fed or ob/ob mice, both analogues had no significant effects on overall blood glucose or plasma insulin levels when compared to saline treated controls. However, desHis¹Glu⁹-glucagon-[mPEG] significantly (P<0.05) annulled glucagon-induced increases in blood glucose and plasma insulin levels in normal mice and had similar non-significant tendencies in high fat and ob/ob mice. In addition, desHis¹Glu⁹(Lys³⁰PAL)-glucagon effectively (P<0.05 to P<0.001) antagonised glucagon-mediated elevations of blood glucose levels in high fat fed and ob/ob mice, but was less efficacious in normal mice. Further studies confirmed the significant persistent glucagon receptor antagonistic properties of both novel enzyme-resistant analogues 4h post administration in normal mice. These studies emphasise the potential of longer-acting peptide-based glucagon receptor antagonists, and particularly acylated versions, for the treatment of diabetes.","is_dataset_classified":null,"base_score":2.833213344056216,"endowment":2.833213344056216,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"23562625","pmcid":null,"openalex_id":"https://openalex.org/W1993170262","authors":[],"funders":[],"total_grants":0,"fwci":1.3293,"citation_percentile":0.80986908,"influential_citations":0,"citation_trend":[{"year":2013,"count":1},{"year":2014,"count":5},{"year":2015,"count":1},{"year":2018,"count":1},{"year":2019,"count":1},{"year":2020,"count":1},{"year":2021,"count":1},{"year":2022,"count":3},{"year":2023,"count":2}],"oa_status":"closed","license":null,"oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0014299913002598?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014299913002598?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ejphar.2013.03.041","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/23562625","host_type":"repository"}],"fields_of_study":["Diabetes Treatment and Management","Pancreatic function and diabetes","Receptor Mechanisms and Signaling","Acylation","Animals","Cell Line","Cyclic AMP","Dipeptidyl Peptidase 4","Drug Stability","Glucagon","HEK293 Cells","Humans","Hyperglycemia","Hyperinsulinism","Hypoglycemic Agents","Insulin","Insulin Secretion","Insulin-Secreting Cells","Male","Mice","Mice, Obese","Obesity","Polyethylene Glycols","Receptors, Glucagon","Recombinant Proteins","Second Messenger Systems"],"mesh_terms":["Insulin Secretion","Acylation","Cyclic AMP","Animals","Cell Line","Drug Stability","Glucagon","Humans","Hyperglycemia","Hyperinsulinism","Hypoglycemic Agents","Insulin","Male","Mice, Obese","Obesity","Polyethylene Glycols","Recombinant Proteins","Second Messenger Systems","Receptors, Glucagon","Dipeptidyl Peptidase 4","Insulin-Secreting Cells","Mice","HEK293 Cells"],"keywords":["Glucagon","Glucagon receptor","Internal medicine","Endocrinology","Glucagon-like peptide 1 receptor","Insulin","Receptor","Chemistry","Glucagon-like peptide-1","Diabetes mellitus","Biology","Medicine","Type 2 diabetes","Agonist"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-17T04:09:22.108565Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
