Cells have multiple repair pathways which attempt to maintain genomic stability. DNA repair proteins have become key targets for therapy, using small molecule inhibitors, in combination with radiation and or chemotherapeutic agents as a means of enhancing cell killing. Significant advances in our understanding of the response of cells to radiation exposures has come from the observation of non-targeted effects where cells respond via mechanisms other than those which are a direct consequence of energy-dependent DNA damage. Typical of these is bystander signalling where cells respond to the fact that their neighbours have been irradiated. Bystander cells show a DNA damage response which is distinct from directly irradiated cells. In bystander cells, ATM- and Rad3-related (ATR) protein kinase-dependent signalling in response to stalled replication forks is an early event in the DNA damage response. The ATM protein kinase is activated downstream of ATR in bystander cells. This offers the potential for differential approaches for the modulation of bystander and direct effects with repair inhibitors which may impact on the response of tumours and on the protection of normal tissues during radiotherapy.","is_dataset_classified":null,"base_score":4.0943445622221,"endowment":4.0943445622221,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"19835868","pmcid":"PMC2855957","openalex_id":"https://openalex.org/W2033577131","authors":[],"funders":[{"funder_name":"Cancer Research UK","grant_id":"A7047","title":null},{"funder_name":"NCI NIH HHS","grant_id":"5P01CA095227-02","title":null},{"funder_name":"CIHR","grant_id":"C1513/A7047","title":null},{"funder_name":"NCI NIH HHS","grant_id":"P01 CA095227","title":null}],"total_grants":4,"fwci":2.0157,"citation_percentile":0.86053956,"influential_citations":0,"citation_trend":[{"year":2012,"count":5},{"year":2013,"count":6},{"year":2014,"count":12},{"year":2015,"count":6},{"year":2016,"count":3},{"year":2017,"count":4},{"year":2018,"count":6},{"year":2019,"count":2},{"year":2021,"count":1},{"year":2022,"count":1},{"year":2023,"count":1},{"year":2024,"count":1}],"oa_status":"green","license":"https://www.elsevier.com/tdm/userlicense/1.0/","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2855957","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2855957","host_type":"repository"},{"url":"https://api.elsevier.com/content/article/PII:S0014299909008917?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0014299909008917?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ejphar.2009.09.068","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/19835868","host_type":"repository"},{"url":"http://www.scopus.com/inward/record.url?scp=71749091838&partnerID=8YFLogxK","host_type":"repository"}],"fields_of_study":["DNA Repair Mechanisms","Effects of Radiation Exposure","PARP inhibition in cancer therapy","Antineoplastic Agents","Bystander Effect","Cell Cycle","Combined Modality Therapy","DNA Breaks, Double-Stranded","DNA Damage","DNA Repair","Humans","Neoplasms","Radiation, Ionizing","Signal Transduction"],"mesh_terms":["Antineoplastic Agents","Cell Cycle","Combined Modality Therapy","DNA Damage","DNA Repair","Humans","Neoplasms","Radiation, Ionizing","Signal Transduction","Bystander Effect","DNA Breaks, Double-Stranded"],"keywords":["Bystander effect","DNA damage","DNA repair","Cell biology","Biology","Protein kinase A","Kinase","Cancer research","Cell","DNA","Genetics","Immunology"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"gen"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-07T22:03:15.024443Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
