Pyrimidine building blocks, in human cells, are synthesised via both de novo biosynthesis and salvage pathways, the latter of which is an effective way of recycling pre-existing nucleotides. As many parasitic organisms lack pyrimidine salvage pathways for pyrimidine nucleotides, blocking de novo biosynthesis is seen as an effective therapeutic means to selectively target the parasite without effecting the human host. Dihydroorotate dehydrogenase (DHODH), which is involved in the de novo biosynthesis of pyrimidines, is a validated target for anti-infective drug research. Recent advances in the DHODH microorganism field are discussed herein, as is the potential for the development of DHODH-targeted therapeutics.","is_dataset_classified":null,"base_score":4.6443908991413725,"endowment":4.6443908991413725,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"31557612","pmcid":null,"openalex_id":"https://openalex.org/W2972679522","authors":[],"funders":[{"funder_name":"Ministero degli Affari Esteri e della Cooperazione Internazionale","grant_id":"PGR00978","title":null},{"funder_name":"Università degli Studi di Torino","grant_id":"BOSD_RILO_17_01","title":null},{"funder_name":"Università degli Studi di Torino","grant_id":"BOSD_RILO_18_01","title":null},{"funder_name":"Università degli Studi di Torino","grant_id":"LOLM_RILO_17_01","title":null},{"funder_name":"Università degli Studi di Torino","grant_id":"LOLM_RILO_18_01","title":null},{"funder_name":"European Medicines Agency","grant_id":"118–122","title":null},{"funder_name":"European Medicines Agency","grant_id":"NCT02856178","title":null},{"funder_name":"European Medicines Agency","grant_id":"NCT03036046","title":null}],"total_grants":8,"fwci":3.4891,"citation_percentile":0.9401472,"influential_citations":0,"citation_trend":[{"year":2020,"count":10},{"year":2021,"count":14},{"year":2022,"count":22},{"year":2023,"count":15},{"year":2024,"count":15},{"year":2025,"count":18},{"year":2026,"count":9}],"oa_status":"green","license":"cc-by-nc-nd","oa_locations":[{"url":"https://www.sciencedirect.com/science/article/pii/S0223523419308311","host_type":"repository"},{"url":"https://www.sciencedirect.com/science/article/pii/S0223523419308311","host_type":"repository"},{"url":"https://api.elsevier.com/content/article/PII:S0223523419308311?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0223523419308311?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ejmech.2019.111681","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/31557612","host_type":"repository"},{"url":"https://doi.org/10.7270/q2mw2mk0","host_type":"repository"}],"fields_of_study":["Biochemical and Molecular Research","Pneumocystis jirovecii pneumonia detection and treatment","HIV/AIDS drug development and treatment"],"mesh_terms":["Dihydroorotate Dehydrogenase","Animals","Anti-Infective Agents","Enzyme Inhibitors","Humans","Plasmodium falciparum","Pyrimidines","Oxidoreductases Acting on CH-CH Group Donors","Drug Discovery"],"keywords":["Dihydroorotate dehydrogenase","Pyrimidine metabolism","Biochemistry","Nucleotide salvage","De novo synthesis","Biosynthesis","Nucleotide","Chemistry","Drug discovery","Dehydrogenase","Antiparasitic agent","Pyrimidine","Enzyme","Biology","Pharmacology","Gene","Purine","Pseudomonas aeruginosa","Malaria","Leishmania","Plasmodium falciparum","Aspergillus fumigatus","Plasmodium vivax","Toxoplasma gondii","Antiviral","Trypanosoma","schistosoma mansoni","Leflunomide","Teriflunomide","Theileria Equi","Babesia Bovis","Babesia Caballi","Bioisosterism","Brequinar","Helicobacter Pilory","Dihydroorotate Dehydrogenase (Dhodh) Inhibitors","Hdhodh"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T15:31:03.264965Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
