However, synthetic pathways often require custom functions or biophysical properties not displayed by natural proteins, limitations that could be overcome through modern protein engineering techniques. Structure-based computational protein design is a powerful tool to engineer new functional capabilities in proteins, and it is beginning to have a profound impact in synthetic biology. Here, we review efforts to increase the capabilities of synthetic biology using computational protein design. We focus primarily on computationally designed proteins not only validated in vitro, but also shown to modulate different activities in living cells. Efforts made to validate computational designs in cells can illustrate both the challenges and opportunities in the intersection of protein design and synthetic biology. We also highlight protein design approaches, which although not validated as conveyors of new cellular function in situ, may have rapid and innovative applications in synthetic biology. We foresee that in the near-future, computational protein design will vastly expand the functional capabilities of synthetic cells.","is_dataset_classified":null,"base_score":0.0,"endowment":0.0,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"29729544","pmcid":null,"openalex_id":null,"authors":[],"funders":[{"funder_name":"European Research Council","grant_id":"716058","title":"Computational Design of Novel Functional Proteins for Immunoengineering"},{"funder_name":"European Commission","grant_id":"665667","title":"Experienced Researchers Incoming Fellowship Programme at the Ecole Polytechnique Fédérale de Lausanne"}],"total_grants":2,"fwci":null,"citation_percentile":null,"influential_citations":0,"citation_trend":[],"oa_status":"bronze","license":"Elsevier TDM","oa_locations":[{"url":"https://www.sciencedirect.com/science/article/am/pii/S0958166917302355","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0958166917302355?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0958166917302355?httpAccept=text/plain","host_type":"publisher"},{"url":"http://infoscience.epfl.ch/record/258553","host_type":"repository"},{"url":"https://zenodo.org/record/3503282","host_type":"repository"},{"url":"https://doi.org/10.1016/j.copbio.2018.04.001","host_type":""},{"url":"https://pubmed.ncbi.nlm.nih.gov/29729544","host_type":""},{"url":"https://dx.doi.org/10.1016/j.copbio.2018.04.001","host_type":""},{"url":"https://sonar.ch/global/documents/180641","host_type":""},{"url":"http://dx.doi.org/10.1016/j.copbio.2018.04.001","host_type":""}],"fields_of_study":["0301 basic medicine","0303 health sciences","03 medical and health sciences"],"mesh_terms":[],"keywords":["Computational Biology","Proteins","Synthetic Biology","Protein Engineering","Enzymes"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-27T18:56:30.738123Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
