However, the impact of upfront D on subsequent therapies is still unexplored. As abiraterone acetate (AA) and enzalutamide (E) are the most commonly used first-line treatment for metastatic castration-resistant prostate cancer (mCRPC), we aimed to assess whether they maintained their efficacy after ADT+D versus ADT alone.
Patients and methods
A cohort of patients with mCRPC treated between 2014 and 2017 with first-line AA or E for mCRPC was identified from 3 hospitals' institutional review board-approved databases. Patients were classified by use of D for mHSPC. This time frame was chosen as ADT+D became a valid therapeutic option for mHSPC in 2014, and it inherently entailed a short follow-up time on AA/E. The endpoints included overall survival from ADT start, overall survival from AA/E start, and time to AA/E start from ADT start. Differences between groups were assessed using the log-rank test.
Results
Of the 102 patients with mCRPC identified, 50 (49%) had previously received ADT alone, while 52 (51%) had ADT+D. No statistically significant difference in any of the evaluated outcomes was observed between the 2 cohorts. Yet, deaths in the ADT+D group were 12 versus 21 in the ADT alone, after a median follow-up of 24.4 and 29.8 months, respectively.
Conclusion
In a cohort of ADT/ADT+D-treated patients with mCRPC with short times to first-line AA/E and follow-up, the efficacy of AA/E is similar regardless of previous use of D.","is_dataset_classified":null,"base_score":2.995732273553991,"endowment":2.995732273553991,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"29331381","pmcid":"PMC5986287","openalex_id":"https://openalex.org/W2776594178","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"P30 CA008748","title":null}],"total_grants":1,"fwci":1.4695,"citation_percentile":0.83390668,"influential_citations":0,"citation_trend":[{"year":2018,"count":1},{"year":2019,"count":2},{"year":2020,"count":5},{"year":2021,"count":5},{"year":2022,"count":2},{"year":2024,"count":3},{"year":2025,"count":1}],"oa_status":"bronze","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"http://www.clinical-genitourinary-cancer.com/article/S1558767317303981/pdf","host_type":"journal"},{"url":"http://www.clinical-genitourinary-cancer.com/article/S1558767317303981/pdf","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1558767317303981?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S1558767317303981?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.clgc.2017.12.012","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/29331381","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/5986287","host_type":"repository"},{"url":"http://hdl.handle.net/2158/1210203","host_type":"repository"}],"fields_of_study":["Prostate Cancer Treatment and Research","Prostate Cancer Diagnosis and Treatment","Cancer, Lipids, and Metabolism","Abiraterone Acetate","Antineoplastic Combined Chemotherapy Protocols","Benzamides","Disease-Free Survival","Docetaxel","Humans","Male","Nitriles","Phenylthiohydantoin","Prostatic Neoplasms, Castration-Resistant","Retrospective Studies","Survival Analysis","Treatment Outcome"],"mesh_terms":["Abiraterone Acetate","Docetaxel","Antineoplastic Combined Chemotherapy Protocols","Benzamides","Humans","Male","Nitriles","Phenylthiohydantoin","Retrospective Studies","Survival Analysis","Treatment Outcome","Disease-Free Survival","Prostatic Neoplasms, Castration-Resistant"],"keywords":["Enzalutamide","Medicine","Prostate cancer","Abiraterone acetate","Docetaxel","Androgen deprivation therapy","Oncology","Internal medicine","Cancer","Androgen receptor","Mcrpc","Chaarted","Ar Targeting Agents","Mcrpc Sequencing","Mhspc"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-02T00:55:05.885578Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
