Here, we report the molecular mechanisms underlying binding of the chromoshadow domain of HP1γ (HP1γ CSD ) to the transcriptional co-repressor KAP1 and HP1γ self-assembly. Using crystallography, NMR, and mass photometry, we show that HP1γ CSD recognizes the HP1 box of KAP1 (KAP1 Hbox ) and forms a relatively stable dimer of dimers, assembled in an antiparallel manner, in contrast to the corresponding HP1α CSD complex, which shows concentration-dependent oligomerization and arrangement of HP1α CSD protomers in a parallel manner. The β-sheet interface between HP1γ CSD dimers is stabilized through electrostatic interactions, unlike the hydrophobic β-sheet interface of HP1α CSD . In vivo rescue experiments using KAP1- and HP1-knockout mouse embryonic stem cells reveal a unique cooperative action of KAP1 and HP1γ, but not other HP1s, in the repression of the long noncoding RNA AI662270, underscoring the notion that cellular functions of HP1 proteins are not redundant.","is_dataset_classified":null,"base_score":0.0,"endowment":0.0,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"41575850","pmcid":"PMC13021124","openalex_id":"https://openalex.org/W7125497045","authors":[],"funders":[{"funder_name":"Welch Foundation","grant_id":"I-2025","title":null},{"funder_name":"Cancer Prevention and Research Institute of Texas","grant_id":"RP250531","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"LE 721/18","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"SFB 1064 Project-ID 213249687","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"425470807","title":null},{"funder_name":"National Institutes of Health","grant_id":"AG067664","title":null},{"funder_name":"National Institutes of Health","grant_id":"GM157928","title":null},{"funder_name":"National Institutes of Health","grant_id":"CA252707","title":null},{"funder_name":"National Institutes of Health","grant_id":"HD109239","title":null},{"funder_name":"National Institutes of Health","grant_id":"GM127020","title":null},{"funder_name":"National Institutes of Health","grant_id":"GM124958","title":null},{"funder_name":"NIA NIH HHS","grant_id":"R01 AG067664","title":null},{"funder_name":"NIGMS NIH HHS","grant_id":"R01 GM157928","title":null},{"funder_name":"NICHD NIH HHS","grant_id":"R01 HD109239","title":null},{"funder_name":"NIGMS NIH HHS","grant_id":"R35 GM124958","title":null},{"funder_name":"NCI NIH HHS","grant_id":"R01 CA252707","title":null},{"funder_name":"NIGMS NIH HHS","grant_id":"R35 GM127020","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"unidentified","title":"unidentified"}],"total_grants":18,"fwci":0.0,"citation_percentile":0.06234824,"influential_citations":0,"citation_trend":[],"oa_status":"gold","license":"cc-by-nc","oa_locations":[{"url":"https://doi.org/10.1016/j.celrep.2025.116874","host_type":"journal"},{"url":"https://doi.org/10.1016/j.celrep.2025.116874","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S2211124725016468?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S2211124725016468?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/41575850","host_type":"repository"},{"url":"https://doaj.org/article/5aeb327dfd844d82abaed587944972f8","host_type":"repository"},{"url":"https://pmc.ncbi.nlm.nih.gov/articles/PMC13021124/","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC13021124","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC13021124?pdf=render","host_type":"Europe_PMC"},{"url":"https://pubmed.ncbi.nlm.nih.gov/41575850/","host_type":""}],"fields_of_study":["Genomics and Chromatin Dynamics","Cancer-related molecular mechanisms research","RNA Research and Splicing","0301 basic medicine","03 medical and health sciences"],"mesh_terms":["Mouse Embryonic Stem Cells","Tripartite Motif-Containing Protein 28","Chromobox Protein Homolog 5","Animals","Chromosomal Proteins, Non-Histone","Humans","Models, Molecular","Protein Binding","Mice","Protein Multimerization","RNA, Long Noncoding"],"keywords":["Antiparallel (mathematics)","Psychological repression","Heterochromatin protein 1","Heterochromatin","Embryonic stem cell","RNA","RNA-binding protein","Transcription (linguistics)","Structure","Mechanism","Lncrna","Kap1","Hp1","Cp: Molecular Biology","Cp: Stem Cell Research","Csd Domain","Article"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"gen"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-12T03:55:02.221652Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
