Among several mechanisms of how EBV may contribute are transcriptome alterations, including changes of antigen processing and preferential presentation of both viral and self-antigens. Here, we report that EBV reprograms the transcriptome and immunopeptidome presented on the MS-associated human leukocyte antigen (HLA)-DR15 molecules of infected B cells. Identical myelin basic protein (MBP) peptides were found to be presented on both EBV-infected B cells and MS brain tissue but not primary B cells and thymic tissue. Peripheral memory and cerebrospinal fluid (CSF)-derived CD4 + T cells of HLA-DR15 + MS patients responded to MBP peptides, MBP (78–90) and/or MBP (83–90) , and T cell clones raised with these peptides recognized all MBP peptides ending at amino acid MBP 90 in MS brain tissue. Our study provides a new mechanistic link for how the environmental and genetic risk factors, EBV infection and HLA-DR15 haplotype, may contribute jointly to MS.","is_dataset_classified":null,"base_score":3.332204510175204,"endowment":3.332204510175204,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"41534530","pmcid":null,"openalex_id":"https://openalex.org/W7123438916","authors":[],"funders":[{"funder_name":"Carl-Zeiss-Stiftung","grant_id":"P2024-02-012","title":null},{"funder_name":"Fundamental Research Funds for the Central Universities","grant_id":"WK3520000016","title":null},{"funder_name":"European Research Council","grant_id":"ERC-2013-ADG 340733","title":null},{"funder_name":"HORIZON EUROPE Framework Programme","grant_id":"101137235","title":"The role of an Epstein-Barr virus infection in the pathogenesis of multiple sclerosis"},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"WA4608/1-2","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"EXC2180-390900677","title":null},{"funder_name":"Deutsche Krebshilfe","grant_id":"70114948","title":null},{"funder_name":"Deutsche Krebshilfe","grant_id":"BW1_4064/03","title":null},{"funder_name":"National Key Research and Development Program of China","grant_id":"2022YFC2304102","title":null},{"funder_name":"Chinese Academy of Sciences","grant_id":"XDB0940303","title":null},{"funder_name":"Chinese Academy of Sciences","grant_id":"XDB0490301","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"92469102","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"82271827","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"82471757","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"32470954","title":null},{"funder_name":"National Natural Science Foundation of China","grant_id":"32300747","title":null},{"funder_name":"Deutsche Forschungsgemeinschaft","grant_id":"unidentified","title":"unidentified"},{"funder_name":"European Commission","grant_id":"340733","title":"Functional Role of the HLA-DR15 Haplotype in Multiple Sclerosis"},{"funder_name":"European Commission","grant_id":"","title":null},{"funder_name":"Deutschen Konsortium für Translationale Krebsforschung","grant_id":"","title":null},{"funder_name":"University of Zurich","grant_id":"","title":null}],"total_grants":21,"fwci":205.9199,"citation_percentile":0.99995641,"influential_citations":0,"citation_trend":[{"year":2026,"count":27}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://doi.org/10.1016/j.cell.2025.12.046","host_type":"journal"},{"url":"https://doi.org/10.1016/j.cell.2025.12.046","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0092867425014953?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0092867425014953?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/41534530","host_type":"repository"},{"url":"http://hdl.handle.net/20.500.11850/793314","host_type":"repository"},{"url":"https://dx.doi.org/10.5167/uzh-290993","host_type":""},{"url":"https://dx.doi.org/10.3929/ethz-c-000793314","host_type":""},{"url":"https://hdl.handle.net/11562/1186787","host_type":""},{"url":"https://doi.org/https://doi.org/10.1016/j.cell.2025.12.046","host_type":""},{"url":"http://dx.doi.org/10.1016/J.CELL.2025.12.046","host_type":""},{"url":"http://dx.doi.org/10.1016/j.cell.2025.12.046","host_type":""}],"fields_of_study":["Multiple Sclerosis Research Studies","vaccines and immunoinformatics approaches","Immunotherapy and Immune Responses","0301 basic medicine","03 medical and health sciences","Humans","Multiple Sclerosis","HLA-DR Serological Subtypes","Herpesvirus 4, Human","Epstein-Barr Virus Infections","Myelin Basic Protein","B-Lymphocytes","Brain","CD4-Positive T-Lymphocytes","Female","Antigen Presentation","Peptides","Adult","Male","Myelin Sheath","Middle Aged"],"mesh_terms":["Adult","B-Lymphocytes","Brain","Myelin Basic Protein","Herpesvirus 4, Human","Female","Humans","Male","Middle Aged","Multiple Sclerosis","Myelin Sheath","Peptides","CD4-Positive T-Lymphocytes","Antigen Presentation","Epstein-Barr Virus Infections","HLA-DR Serological Subtypes"],"keywords":["Multiple sclerosis","Transcriptome","Antigen presentation","Myelin","Antigen","Antigen processing","Myelin basic protein","Peptide","Brain tissue","Epstein-Barr virus","B cells","Immunopeptidome","Thymic Tissue","Autoreactive Cd4(+) T Cells","Myelin Basic Protein Peptide","Male","CD4-Positive T-Lymphocytes","Adult","Epstein-Barr Virus Infections","Herpesvirus 4, Human","Humans","610 Medicine &
