{"doi":"10.1016/j.cdnut.2023.100501","title":"P13-045-23 Combining Gamma-Tocopherol (γT) and Aspirin Synergistically Suppresses Colitis-Associated Colon Tumorigenesis and Modulates Gut Microbiota in Mice","abstract":"Objectives: Our objective is to explore the combinatory effects of gamma-tocopherol(T) and aspirin on the development of colitis-associated colorectal cancer (CAC).We hypothesize that combining T and aspirin is better than either compound singly for suppressing CAC.Methods: AIN93G diets supplemented with aspirin (250 ppm), or T (500 ppm) or their combination were given to AOM/DSS treated male Balb/c mice for 9 weeks.Histopathological analysis and multiplex-based immunoassay of cytokines were conducted using colon tissues collected.Gut microbiome were analyzed based 16S rRNA sequencing followed by QIIME2 analysis using feces collected.Cell viability was examined using MTT assay.Results: Compared to the control, combining aspirin and T but not either compound alone significantly reduced AOM/DSSinduced tumor area and multiplicity of large-size tumors by 60% and 50%, respectively.Meanwhile, T mitigated aspirin-promoted inflammation and stomach lesions in mice.Moreover, the combination appeared to cause favorable changes of gut microbiota compared to the control and synergistically suppressed the growth of HCT116 cells.Conclusions: Our study demonstrates that combining aspirin and T improves anticancer effects and counteracted side effects compared to aspirin and may therefore be a novel combinatory chemopreventive agent against CAC.","journal":"Current Developments in Nutrition","year":2023,"id":409903,"datarank":0.0,"base_score":0.0,"endowment":0.0,"self_citation_contribution":0.0,"citation_network_contribution":0.0,"self_endowment_contribution":0.0,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":0,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":0.962,"is_data_producer":false,"deposit_databanks":null,"is_oa":true,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":"2023-01-01","fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":632774,"name":"Qing Jiang","orcid":"0000-0002-0579-8280","position":1,"is_corresponding":false},{"id":486924,"name":"Yava Jones‐Hall","orcid":"0000-0003-4030-9830","position":2,"is_corresponding":false},{"id":684132,"name":"Kilia Y.
