Mammals also have 5-hydroxymethylcytosine (5hmC), resulting from hydroxylation of 5mC by TET family-enzymes. 5hmC is abundant in mouse Purkinje neurons and embryonic stem cells, and regarded as an important intermediate for active DNA demethylation in mammals. However, the presence of 5hmC in plants has not been clearly demonstrated. In Arabidopsis, the DEMETER (DME) family DNA glycosylases efficiently remove 5mC, which results in DNA demethylation and transcriptional activation of target genes. Here we show that DME and ROS1 have a significant 5hmC excision activity in vitro, although we detected no 5hmC in Arabidopsis, suggesting that it is very unlikely for plants to utilize 5hmC as a DNA demethylation intermediate. Our results indicate that both plants and animals have 5mC in common but DNA demethylation systems have independently evolved with distinct mechanisms.","is_dataset_classified":null,"base_score":3.6888794541139363,"endowment":3.6888794541139363,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"24661881","pmcid":"PMC5028334","openalex_id":"https://openalex.org/W2026388592","authors":[],"funders":[{"funder_name":"Rural Development Administration","grant_id":"NRF-2013H1A2A1034385","title":null},{"funder_name":"NIEHS NIH HHS","grant_id":"R01 ES019625","title":null},{"funder_name":"National Research Foundation of Korea","grant_id":"","title":null}],"total_grants":3,"fwci":2.0796,"citation_percentile":0.86985347,"influential_citations":0,"citation_trend":[{"year":2014,"count":4},{"year":2015,"count":5},{"year":2016,"count":3},{"year":2017,"count":4},{"year":2018,"count":2},{"year":2019,"count":3},{"year":2020,"count":3},{"year":2021,"count":2},{"year":2022,"count":5},{"year":2023,"count":5},{"year":2024,"count":2},{"year":2026,"count":1}],"oa_status":"closed","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0006291X14005038?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0006291X14005038?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.bbrc.2014.03.060","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/24661881","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/5028334","host_type":"repository"}],"fields_of_study":["Epigenetics and DNA Methylation","Genomics and Chromatin Dynamics","5-Methylcytosine","Animals","Arabidopsis","Arabidopsis Proteins","Cytosine","DNA Glycosylases","DNA Methylation","Mice","N-Glycosyl Hydrolases","Nuclear Proteins","Signal Transduction","Trans-Activators"],"mesh_terms":["Animals","Cytosine","Nuclear Proteins","N-Glycosyl Hydrolases","Signal Transduction","Trans-Activators","Arabidopsis","DNA Methylation","Arabidopsis Proteins","5-Methylcytosine","DNA Glycosylases","Mice"],"keywords":["DNA demethylation","5-Hydroxymethylcytosine","DNA glycosylase","DNA methylation","Biology","Epigenetics","5-Methylcytosine","Epigenomics","DNA","Arabidopsis","Transposable element","Cell biology","Gene","Genetics","DNA repair","Gene expression","Genome","Mutant","Demeter"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-26T13:33:38.565165Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
