Research has predominantly focused on how oncogenes promote the development of different TC subtypes by influencing the downstream signaling pathways. Targeted therapies show significant efficacy; however, they often induce drug resistance through feedback activation or compensatory signaling bypasses. Recent evidence indicates that thyroid oncogenes initiate and mediate TC progression, and contribute to drug resistance in distinct TC subtypes through induced metabolic reprogramming and immune microenvironment remodeling. Hence, we propose the concept \"Oncogene-Metabolism-Immunity axis.\" We discussed the molecular mechanisms by which oncogene-driven metabolic reprogramming and tumor immune microenvironment Remodeling (TIME), and their mutual interactions, induce TC progression, drug resistance, and immune evasion. Finally, we systematically evaluated and summarized potential strategies targeting key oncogenes, metabolic catalysts, immune checkpoints (ICs), and combination therapies to enhance the efficacy of targeted treatments for TC and overcome drug resistance.","is_dataset_classified":null,"base_score":1.0986122886681096,"endowment":1.0986122886681096,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"41005624","pmcid":null,"openalex_id":"https://openalex.org/W4414521747","authors":[],"funders":[{"funder_name":"Lanzhou University","grant_id":"Ldyyyn2021\\u201030","title":null},{"funder_name":"Natural Science Foundation of Gansu Province","grant_id":"17JR5RA272","title":null},{"funder_name":"Natural Science Foundation of Gansu Province","grant_id":"22JR5RA923","title":null}],"total_grants":3,"fwci":0.9423,"citation_percentile":0.76580976,"influential_citations":0,"citation_trend":[{"year":2026,"count":2}],"oa_status":"hybrid","license":"cc-by-nc-nd","oa_locations":[{"url":"https://doi.org/10.1016/j.bbcan.2025.189459","host_type":"journal"},{"url":"https://doi.org/10.1016/j.bbcan.2025.189459","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0304419X2500201X?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0304419X2500201X?httpAccept=text/plain","host_type":"publisher"},{"url":"https://pubmed.ncbi.nlm.nih.gov/41005624","host_type":"repository"}],"fields_of_study":["Cancer, Hypoxia, and Metabolism","Thyroid Cancer Diagnosis and Treatment","Ferroptosis and cancer prognosis"],"mesh_terms":["Animals","Humans","Mutation","Oncogenes","Thyroid Neoplasms","Signal Transduction","Drug Resistance, Neoplasm","Precision Medicine","Tumor Microenvironment"],"keywords":["Reprogramming","Immune system","Tumor microenvironment","Thyroid cancer","Drug resistance","Cancer","Drug development","Drug","Signal transduction","Combination therapy","Targeted Therapy","Metabolic Reprogramming","Immune Microenvironment Remodeling","Carcinogenic Mutations"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T15:21:21.670045Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
