Two shared nucleotide-binding sites (NBS) participate in this cycle. In asymmetric ABC pumps, only one of them hydrolyzes ATP, and the functional role of the other remains unclear. Using a drug-based selection strategy on the transport-deficient mutant L529A in the transmembrane domain of the Candida albicans pump Cdr1p; we identified a spontaneous secondary mutation restoring drug-translocation. The compensatory mutation Q1005H was mapped 60 Å away, precisely in the ABC signature sequence of the non-hydrolytic NBS. The same was observed in the homolog Cdr2p. Both the mutant and suppressor proteins remained ATPase active, but remarkably, the single Q1005H mutant displayed a two-fold reduced ATPase activity and a two-fold increased drug-resistance as compared to the wild-type protein, pointing at a direct control of the non-hydrolytic NBS in substrate-translocation through ATP binding in asymmetric ABC pumps.","is_dataset_classified":null,"base_score":2.995732273553991,"endowment":2.995732273553991,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"31734312","pmcid":null,"openalex_id":"https://openalex.org/W2984740793","authors":[],"funders":[{"funder_name":"Department of Biotechnology, Ministry of Science and Technology","grant_id":"","title":null},{"funder_name":"Agence Nationale de la Recherche","grant_id":"","title":null}],"total_grants":2,"fwci":0.7157,"citation_percentile":0.72526131,"influential_citations":0,"citation_trend":[{"year":2020,"count":1},{"year":2021,"count":2},{"year":2022,"count":5},{"year":2023,"count":6},{"year":2024,"count":1},{"year":2025,"count":4}],"oa_status":"closed","license":"http://www.elsevier.com/open-access/userlicense/1.0/","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0005273619302792?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0005273619302792?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.bbamem.2019.183131","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/31734312","host_type":"repository"},{"url":"https://hal.science/hal-02376201","host_type":"repository"}],"fields_of_study":["Drug Transport and Resistance Mechanisms","Antibiotic Resistance in Bacteria","RNA Interference and Gene Delivery","ATP-Binding Cassette Transporters","Adenosine Triphosphate","Antifungal Agents","Binding Sites","Candida albicans","Drug Resistance, Fungal","Fungal Proteins","Mutation","Protein Binding"],"mesh_terms":["Adenosine Triphosphate","Antifungal Agents","Binding Sites","Candida albicans","Fungal Proteins","Mutation","Protein Binding","ATP-Binding Cassette Transporters","Drug Resistance, Fungal"],"keywords":["ATP-binding cassette transporter","ATP hydrolysis","Chromosomal translocation","Mutant","Cyclic nucleotide-binding domain","Transmembrane domain","Biochemistry","ATPase","Candida albicans","Walker motifs","Chemistry","Transport protein","Biology","Cell biology","Nucleotide","Transporter","Enzyme","Genetics","Gene","ABC transporter","Antifungal Drug Resistance","Abc Signature Sequence","Non-hydrolytic Nucleotide-binding Site"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-17T20:21:09.719512Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
