Wnt signaling is mediated by frizzled receptors (Fzds) at the cell surface and can be modulated by the secreted frizzled-related proteins (SFRPs) and other molecular antagonists. Abnormal Wnt signaling has been implicated in several diseases. However, due to the complexity of the Wnt signal and the lack of knowledge pertaining to the binding properties of different Wnt ligands, no therapeutic agents that target this pathway exist. Using a novel enzyme-linked immunosorbent assay (ELISA)-based technique, we were able to determine the first measurements of binding affinity for specific Wnt interactions. This study shows that purified Wnt3a, Wnt7a, and Wnt5a have different binding specificities for Fzds and SFRPs.","is_dataset_classified":null,"base_score":3.6888794541139363,"endowment":3.6888794541139363,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"20227380","pmcid":"PMC2881539","openalex_id":"https://openalex.org/W2081949986","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"P50 CA098258","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"R90 DK071505","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"T90 DK070109","title":null},{"funder_name":"NCI NIH HHS","grant_id":"P50-CA098258","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"5 R90 DK071505-03","title":null},{"funder_name":"NIDDK NIH HHS","grant_id":"5 T 90 DK070109-03","title":null}],"total_grants":6,"fwci":2.5507,"citation_percentile":0.89486346,"influential_citations":0,"citation_trend":[{"year":2012,"count":8},{"year":2013,"count":8},{"year":2014,"count":2},{"year":2015,"count":2},{"year":2016,"count":2},{"year":2017,"count":4},{"year":2018,"count":1},{"year":2019,"count":2},{"year":2020,"count":1},{"year":2021,"count":2},{"year":2022,"count":1},{"year":2023,"count":1},{"year":2025,"count":2}],"oa_status":"closed","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0003269710001612?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0003269710001612?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ab.2010.03.009","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/20227380","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/2881539","host_type":"repository"}],"fields_of_study":["Wnt/β-catenin signaling in development and cancer","Cancer-related gene regulation"],"mesh_terms":["Secreted Frizzled-Related Proteins","Enzyme-Linked Immunosorbent Assay","Female","Humans","Protein Binding","Proto-Oncogene Proteins","Endometrial Neoplasms","Receptors, G-Protein-Coupled","Cell Line, Tumor","MAP Kinase Kinase 4","Wnt Proteins","Frizzled Receptors","beta Catenin","Wnt3 Protein","Wnt3A Protein"],"keywords":["Frizzled","Wnt signaling pathway","WNT3A","Receptor","Biology","LRP6","Affinities","Cell biology","Binding affinities","Glycoprotein","Signal transduction","LRP5","Computational biology","Biochemistry"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-25T01:42:26.691386Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
