Inhibition of bacterial ligase is expected to cause chromosome degradation and cell death, making it an attractive target for new antibacterials. The prototypical bacterial ligase couples the hydrolysis of NAD(+) to phosphodiester bond formation between an adjacent 3'OH and 5'-terminal phosphate of nicked duplex DNA. The first step is the reversible formation of a ligase-adenylate from the reaction between apoenzyme and NAD(+). Inhibitors that compete with NAD(+) are expected to be bacterial specific because eukaryotic DNA ligases use ATP and differ in the sequence composition of their adenylation domain. We report here a high-throughput assay that measures the adenylation reaction specifically by monitoring ligase-AMP formation via scintillation proximity technologies. Escherichia coli DNA ligase was biotinylated in vivo; after reaction with radiolabeled NAD(+), ligase-[(3)H]AMP could be captured onto the streptavidin-coated surface of the solid scintillant. The method was ideal for high-throughput screening because it required minimal manipulations and generated a robust signal with minimal scatter. Certain adenosine analogs were found to inhibit the adenylation assay and had similar potency of inhibition in a DNA ligation assay.","is_dataset_classified":null,"base_score":2.833213344056216,"endowment":2.833213344056216,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"17493575","pmcid":null,"openalex_id":"https://openalex.org/W2013087839","authors":[],"funders":[],"total_grants":0,"fwci":0.4882,"citation_percentile":0.57744694,"influential_citations":0,"citation_trend":[{"year":2012,"count":4},{"year":2013,"count":1},{"year":2014,"count":1},{"year":2015,"count":1},{"year":2016,"count":1},{"year":2018,"count":1},{"year":2019,"count":2},{"year":2020,"count":1}],"oa_status":"closed","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S0003269707001911?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S0003269707001911?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1016/j.ab.2007.03.028","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/17493575","host_type":"repository"}],"fields_of_study":["Antibiotic Resistance in Bacteria","Advanced biosensing and bioanalysis techniques","DNA and Nucleic Acid Chemistry","Adenosine","Adenosine Monophosphate","Adenosine Triphosphate","Base Sequence","Biotin","Chemistry Techniques, Analytical","DNA","DNA Ligases","Enzyme Inhibitors","Escherichia coli","Kinetics","Ligands","NAD","Reproducibility of Results","Scintillation Counting","Streptavidin"],"mesh_terms":["Adenosine","Adenosine Monophosphate","Adenosine Triphosphate","Base Sequence","Biotin","Chemistry Techniques, Analytical","DNA","Enzyme Inhibitors","Escherichia coli","Kinetics","Ligands","NAD","DNA Ligases","Scintillation Counting","Reproducibility of Results","Streptavidin"],"keywords":["DNA ligase","Okazaki fragments","Phosphodiester bond","DNA Ligases","Adenylylation","Biochemistry","NAD+ kinase","Biology","DNA","Biotinylation","DNA replication","Ligase chain reaction","Ubiquitin ligase","DNA polymerase I","In vitro recombination","Molecular biology","DNA repair","Sequencing by ligation","Enzyme","Ubiquitin","Eukaryotic DNA replication","Genomic library","RNA","Biosynthesis","Polymerase chain reaction","Complementary DNA","Molecular cloning"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T19:20:33.552427Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
