Here, we analyze recent myeloma RCTs and evaluate the issues relevant to current and future myeloma RCT design.<h4>Recent findings</h4>A review of recent phase III RCTs that led to approval of new agents/combinations reveals that none were designed with OS as the primary endpoint, but instead utilized time to progression (TTP) or progression-free survival (PFS). These studies illuminate the inherent difficulties of designing trials with the primary endpoint of OS/PFS in a disease characterized by increasingly prolonged survival times, availability of effective salvage therapies, and competing events such as co-morbid conditions. Alternative primary endpoints other than OS or PFS need to be developed for future myeloma RCTs. Validated surrogate endpoints with novel clinical trial designs will help improve the feasibility of conducting comparative clinical trials in a timely manner.","is_dataset_classified":null,"base_score":3.4011973816621555,"endowment":3.4011973816621555,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"30661162","pmcid":"PMC9344397","openalex_id":"https://openalex.org/W2910926762","authors":[],"funders":[{"funder_name":"NCI NIH HHS","grant_id":"P30 CA016056","title":null}],"total_grants":1,"fwci":2.0936,"citation_percentile":0.87368741,"influential_citations":0,"citation_trend":[{"year":2018,"count":1},{"year":2019,"count":2},{"year":2020,"count":2},{"year":2021,"count":3},{"year":2022,"count":8},{"year":2023,"count":4},{"year":2024,"count":4},{"year":2025,"count":3},{"year":2026,"count":2}],"oa_status":"green","license":"http://www.springer.com/tdm","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/9344397","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/9344397","host_type":"repository"},{"url":"http://link.springer.com/content/pdf/10.1007/s11899-019-0495-9.pdf","host_type":"publisher"},{"url":"http://link.springer.com/article/10.1007/s11899-019-0495-9/fulltext.html","host_type":"publisher"},{"url":"https://doi.org/10.1007/s11899-019-0495-9","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/30661162","host_type":"repository"}],"fields_of_study":["Multiple Myeloma Research and Treatments","Protein Degradation and Inhibitors","Histone Deacetylase Inhibitors Research","Humans","Multiple Myeloma","Survival Rate"],"mesh_terms":["Humans","Multiple Myeloma","Survival Rate"],"keywords":["Clinical endpoint","Medicine","Multiple myeloma","Surrogate endpoint","Clinical trial","Randomized controlled trial","Progression-free survival","Oncology","Internal medicine","Hematology","Overall survival","Clinical study design","Intensive care medicine","Primary Endpoint"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"nct"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-08T01:34:27.265287Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
