The protease inhibitors TLCK, TPCK, PMSF, MG-132 and PSI markedly inhibited DNA synthesis. The inhibitory effects were concentration-dependent and of early onset (within 60 min). The most selective proteasome inhibitors lactacystin and clasto-lactacystin-beta-lactone as well as the calpain inhibitor I and II had no or minimal effects on DNA synthesis. Only high concentrations of calpain inhibitor I (>or= 250 microM) and calpain inhibitor II (>or= 500 microM) gave a DNA synthesis inhibition. These results suggest that (1) ongoing DNA replication is regulated by proteolysis and (2) the proteolytic pathways involved are neither the proteasome nor the calpains.","is_dataset_classified":null,"base_score":1.0986122886681096,"endowment":1.0986122886681096,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"19288291","pmcid":null,"openalex_id":"https://openalex.org/W2183313302","authors":[],"funders":[],"total_grants":0,"fwci":0.0,"citation_percentile":0.20773832,"influential_citations":0,"citation_trend":[{"year":2014,"count":1},{"year":2016,"count":1}],"oa_status":"closed","license":"https://www.springernature.com/gp/researchers/text-and-data-mining","oa_locations":[{"url":"https://link.springer.com/content/pdf/10.1007/s10637-009-9238-4.pdf","host_type":"publisher"},{"url":"https://link.springer.com/article/10.1007/s10637-009-9238-4/fulltext.html","host_type":"publisher"},{"url":"http://link.springer.com/content/pdf/10.1007/s10637-009-9238-4","host_type":"publisher"},{"url":"https://doi.org/10.1007/s10637-009-9238-4","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/19288291","host_type":"repository"}],"fields_of_study":["Calpain Protease Function and Regulation","Ubiquitin and proteasome pathways","Cell death mechanisms and regulation","Medicine","Biology","Animals","Animals, Newborn","Cerebral Cortex","Cysteine Proteinase Inhibitors","DNA","Dose-Response Relationship, Drug","In Vitro Techniques","Nucleic Acid Synthesis Inhibitors","Proteasome Endopeptidase Complex","Protein Synthesis Inhibitors","Rats","Rats, Sprague-Dawley","Serine Proteinase Inhibitors","Signal Transduction"],"mesh_terms":["Animals","Animals, Newborn","Cerebral Cortex","DNA","Dose-Response Relationship, Drug","Protein Synthesis Inhibitors","Signal Transduction","Serine Proteinase Inhibitors","Cysteine Proteinase Inhibitors","Rats, Sprague-Dawley","Nucleic Acid Synthesis Inhibitors","Proteasome Endopeptidase Complex","Rats","In Vitro Techniques"],"keywords":["Calpain","Proteolysis","Proteasome","Cell biology","Cerebral cortex","DNA","Chemistry","Biology","Neuroscience","Biochemistry","Molecular biology","Enzyme"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-16T04:51:26.837074Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
