While the two proteins display strong structural conservation and the mechanisms underlying membrane fusion are similar, they share no sequence similarity. Whether they are functionally interchangeable is currently unknown. In this study, we constructed scIAV-S, a single-cycle influenza A virus pseudotyped with the spike protein of porcine epidemic diarrhea virus (PEDV), and demonstrated that this virus could infect cultured cells and trigger massive syncytium formation. Treatment with endocytosis inhibitors did not affect syncytium formation by infected cells. Moreover, the infectivity of scIAV-S was associated with the degree of cell adaptation of PEDV-S. Intriguingly, scIAV-S lacking functional neuraminidase (NA) exhibited substantially higher infectivity, suggesting a pivotal role of the sialic acid in the binding/entry of PEDV. Together, scIAV-S offers a robust platform for the investigation of the entry mechanism of PEDV or, possibly, of other coronaviruses.","is_dataset_classified":null,"base_score":1.791759469228055,"endowment":1.791759469228055,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"30136251","pmcid":"PMC7087185","openalex_id":"https://openalex.org/W2888659845","authors":[],"funders":[{"funder_name":"National Science and Technology Development Agency","grant_id":"P15-51261","title":null}],"total_grants":1,"fwci":0.4077,"citation_percentile":0.61614486,"influential_citations":0,"citation_trend":[{"year":2018,"count":1},{"year":2019,"count":1},{"year":2023,"count":2},{"year":2025,"count":1}],"oa_status":"closed","license":"https://www.springernature.com/gp/researchers/text-and-data-mining","oa_locations":[{"url":"https://link.springer.com/article/10.1007/s00705-018-4001-9/fulltext.html","host_type":"publisher"},{"url":"https://link.springer.com/content/pdf/10.1007/s00705-018-4001-9.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1007/s00705-018-4001-9","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/30136251","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/7087185","host_type":"repository"},{"url":"https://europepmc.org/articles/PMC7087185","host_type":"Europe_PMC"},{"url":"https://europepmc.org/articles/PMC7087185?pdf=render","host_type":"Europe_PMC"}],"fields_of_study":["Animal Virus Infections Studies","Virus-based gene therapy research","Viral gastroenteritis research and epidemiology"],"mesh_terms":["Animals","Cell Line","Influenza A virus","Recombinant Fusion Proteins","Swine","Swine Diseases","Coronavirus Infections","Porcine epidemic diarrhea virus","Spike Glycoprotein, Coronavirus"],"keywords":["Biology","Virology","Syncytium","Porcine epidemic diarrhea virus","Infectivity","Virus","Lipid bilayer fusion","Coronavirus","Viral entry","Hemagglutinin (influenza)","Neuraminidase","Cell fusion","Viral envelope","Vero cell","Glycoprotein","Cell culture","Viral replication","Molecular biology","Genetics"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[{"name":"gen"}],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-03T10:53:55.306902Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
