In this study we investigated the relative role of different pathways in human skin fibroblast proliferation. Using chemical inhibitors and activators of signal transduction, we demonstrated that: (i) AC/cAMP and PLC/1,4,5 inositol triphosphate/diacylglycerol second-messenger systems are simultaneously activated following PTHrP binding to its receptors; (ii) the mitogenic response to PTHrP derives from a balance between two counteracting pathways--an activating route mediated by protein kinase C (PKC) and an inhibitory route mediated by protein kinase A (PKA); (iii) PTHrP mitogenic effects are largely dependent on MAPKs, whose activity can be modulated by both PKA and PKC. Our results indicate that MAPKs are common targets of both transduction routes and, at the same time, their point of divergence in mediating PTHrP dual and opposite mitogenic effects.","is_dataset_classified":null,"base_score":3.258096538021482,"endowment":3.258096538021482,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"12568342","pmcid":"PMC11146128","openalex_id":"https://openalex.org/W2065091962","authors":[],"funders":[],"total_grants":0,"fwci":0.547,"citation_percentile":0.63514931,"influential_citations":0,"citation_trend":[{"year":2012,"count":1},{"year":2013,"count":1},{"year":2014,"count":3},{"year":2016,"count":1},{"year":2017,"count":1},{"year":2025,"count":2}],"oa_status":"green","license":"http://www.springer.com/tdm","oa_locations":[{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/11146128","host_type":"repository"},{"url":"https://www.ncbi.nlm.nih.gov/pmc/articles/11146128","host_type":"repository"},{"url":"http://link.springer.com/content/pdf/10.1007/s000180200015.pdf","host_type":"publisher"},{"url":"http://link.springer.com/article/10.1007/s000180200015/fulltext.html","host_type":"publisher"},{"url":"http://link.springer.com/content/pdf/10.1007/s000180200015","host_type":"publisher"},{"url":"https://doi.org/10.1007/s000180200015","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/12568342","host_type":"repository"},{"url":"http://hdl.handle.net/11392/1516141","host_type":"repository"},{"url":"http://hdl.handle.net/11365/21281","host_type":"repository"}],"fields_of_study":["Protein Kinase Regulation and GTPase Signaling","Bone health and treatments","Melanoma and MAPK Pathways","Adult","Cells, Cultured","Colforsin","Cyclic AMP","Cyclic AMP-Dependent Protein Kinases","Dermis","Enzyme Activation","Enzyme Inhibitors","Fibroblasts","Humans","Mitogen-Activated Protein Kinases","Parathyroid Hormone-Related Protein","Peptide Hormones","Protein Binding","Protein Kinase C","Receptor, Parathyroid Hormone, Type 1","Receptors, Parathyroid Hormone","Second Messenger Systems"],"mesh_terms":["Cyclic AMP","Adult","Cells, Cultured","Enzyme Activation","Enzyme Inhibitors","Fibroblasts","Colforsin","Humans","Protein Binding","Protein Kinase C","Second Messenger Systems","Cyclic AMP-Dependent Protein Kinases","Receptors, Parathyroid Hormone","Dermis","Mitogen-Activated Protein Kinases","Peptide Hormones","Parathyroid Hormone-Related Protein","Receptor, Parathyroid Hormone, Type 1"],"keywords":["Protein kinase C","Diacylglycerol kinase","Second messenger system","Signal transduction","Gq alpha subunit","Protein kinase A","Cell biology","Phospholipase C","Receptor","Inositol phosphate","MAPK/ERK pathway","G protein","Kinase","Chemistry","Biology","Inositol","Biochemistry"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T12:04:27.129647Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
