{"doi":"10.1007/978-3-031-40949-3_18","title":"Drug Delivery Across the Intact Tympanic Membrane: Methods, Mechanisms and Potential Impact","abstract":"The common occurrence of middle ear ailments such as otitis media has spurred research efforts to enhance options for active delivery of drugs and other therapies through the intact tympanic membrane (TM). Utilizing bacteriophage display in the rat, we identified a rare set of peptides that are able to transit phage particles placed on the surface of the intact TM into the middle ear cavity. This trans-tympanic transport occurs through an active process and appears not to cause damage to middle or inner ear function. The peptides support transport across the TM in other species, including humans. Trans-tympanic drug delivery could achieve higher middle ear drug levels and avoid side- or off-target effects when compared to systemic medications.
