Initially described to deliver intracellular antigens to MHC class II loading compartments, its molecular machinery has now also been described to impact the delivery of extracellular antigens to MHC class II loading compartments through the noncanonical use of the macroautophagy machinery during LC3-associated phagocytosis (LAP). Therefore, in pathological situations (viral or bacterial infections, tumorigenesis) the pathway might be involved in shaping CD4<sup>+</sup> T cell responses.In this chapter we describe three basic experiments for the monitoring and manipulation of macroautophagic antigen processing toward MHC class II presentation through the canonical pathway. Firstly, we will discuss how to monitor autophagic flux and autophagosome fusion with MHC class II loading compartments. Secondly, we will show how to target proteins to autophagosomes in order to monitor macroautophagy dependent antigen processing via their enhanced presentation on MHC class II molecules to CD4<sup>+</sup> T cells. And finally, we will describe how macroautophagy can be silenced in antigen presenting cells, like human monocyte-derived dendritic cells (DCs).","is_dataset_classified":null,"base_score":0.0,"endowment":0.0,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"31147952","pmcid":null,"openalex_id":null,"authors":[],"funders":[{"funder_name":"Swiss National Science Foundation","grant_id":"160708","title":"Identify new molecular mechanisms and cellular precursors to augment T cell function in chronic infections and tumors"}],"total_grants":1,"fwci":null,"citation_percentile":null,"influential_citations":0,"citation_trend":[],"oa_status":"green","license":"other-oa","oa_locations":[{"url":"http://link.springer.com/content/pdf/10.1007/978-1-4939-9450-2_25","host_type":"publisher"},{"url":"https://doi.org/10.5167/uzh-183067","host_type":"repository"},{"url":"https://doi.org/10.1007/978-1-4939-9450-2_25","host_type":""},{"url":"https://www.zora.uzh.ch/id/eprint/183067/1/monitoring_antigen_processing_for_mhc_presentation_via_macroautophagy_final_manuscript.pdf","host_type":""},{"url":"https://dx.doi.org/10.5167/uzh-183067","host_type":""},{"url":"https://pubmed.ncbi.nlm.nih.gov/31147952","host_type":""},{"url":"https://dx.doi.org/10.1007/978-1-4939-9450-2_25","host_type":""},{"url":"https://sonar.ch/global/documents/258415","host_type":""},{"url":"https://www.zora.uzh.ch/id/eprint/183067/","host_type":""}],"fields_of_study":["0301 basic medicine","03 medical and health sciences","0303 health sciences"],"mesh_terms":["Dendritic Cells","CD4-Positive T-Lymphocytes","Monocytes","Clone Cells","Humans","Green Fluorescent Proteins","Membrane Proteins","Antigens, Neoplasm","Histocompatibility Antigens Class II","Immunologic Techniques","Antigen Presentation","Interferon-gamma","HEK293 Cells","A549 Cells","Autophagosomes","Macroautophagy"],"keywords":["Cd4+ T Cells","Macroautophagy","Mhc Class Ii Loading Compartment","CD4-Positive T-Lymphocytes","Green Fluorescent Proteins","610 Medicine & health","10263 Institute of Experimental Immunology","Monocytes","Interferon-gamma","1311 Genetics","Antigens, Neoplasm","1312 Molecular Biology","Humans","610 Medicine & health","Antigen Presentation","Autophagosomes","Histocompatibility Antigens Class II","Membrane Proteins","Dendritic Cells","Clone Cells","HEK293 Cells","A549 Cells","Immunologic Techniques","570 Life sciences; biology"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-27T17:30:51.511770Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
