However, cell culture protocols for iPSC reprogramming, neural induction and differentiation tend to be labor and time intensive, costly and commonly depend on viral vector delivery. Single-step reprogramming to induced neural stem cells (iNSC) avoids many of the necessary intermediate steps of the aforementioned method but yields a cell type that proliferates over longer time spans and readily differentiates to mature neurons when required. Here we describe a plasmid based reprogramming protocol employing defined, commercially available components for induction and proliferation of iNSC, followed by a defined, small molecule based differentiation step toward mature neurons. The described method might be of particular interest for groups with limited resources and/or restricted access to higher biosafety level facilities required for viral transduction, but also for groups requiring a high throughput for dealing with large numbers of cell lines.","is_dataset_classified":null,"base_score":1.791759469228055,"endowment":1.791759469228055,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"30196399","pmcid":null,"openalex_id":"https://openalex.org/W2891534580","authors":[],"funders":[],"total_grants":0,"fwci":1.5894,"citation_percentile":0.8329148,"influential_citations":0,"citation_trend":[{"year":2020,"count":4},{"year":2024,"count":1}],"oa_status":"closed","license":"http://www.springer.com/tdm","oa_locations":[{"url":"http://link.springer.com/content/pdf/10.1007/978-1-4939-8697-2_2","host_type":"publisher"},{"url":"https://doi.org/10.1007/978-1-4939-8697-2_2","host_type":"book series"},{"url":"https://pubmed.ncbi.nlm.nih.gov/30196399","host_type":"repository"}],"fields_of_study":["Pluripotent Stem Cells Research","CRISPR and Genetic Engineering","Biomedical Ethics and Regulation","Cell Culture Techniques","Cell Differentiation","Cells, Cultured","Cellular Reprogramming","Colony-Forming Units Assay","Dermis","Fibroblasts","Fluorescent Antibody Technique","Gene Expression","Humans","Neural Stem Cells","Neurons","Plasmids","Transfection","Transgenes"],"mesh_terms":["Cell Differentiation","Cells, Cultured","Colony-Forming Units Assay","Fibroblasts","Fluorescent Antibody Technique","Humans","Neurons","Plasmids","Transfection","Gene Expression","Cell Culture Techniques","Transgenes","Dermis","Neural Stem Cells","Cellular Reprogramming"],"keywords":["Reprogramming","Induced pluripotent stem cell","Somatic cell","Biology","Cell biology","Neural stem cell","Regenerative medicine","Transduction (biophysics)","Stem cell","Cellular differentiation","Viral vector","Cell type","Cell culture","Cell","Computational biology","Embryonic stem cell","Genetics","Gene","Biochemistry","Small molecules","plasmid transfection","Human Dermal Fibroblasts","Induced Neural Stem Cells","Human Neurons","Direct Neural Reprogramming","Small Molecule Based Differentiation"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Decent work and economic growth"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-07-28T18:32:03.177283Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
