Fas cross-linking resulted in a both dose- and time-dependent increases in phospholipase D activity. There was a nearly maximum saturated rise in phospholipase D activity at the dose of 200 ng/ml anti-Fas monoclonal antibody showing a fourfold increase within 3 h. Fas activation also caused an approximately twofold increase of phosphatidylcholine-specific phospholipase C activity and 1,2-diacylglycerol release, which could be blocked by 30 min pretreatment with the phosphatidylcholine-specific phospholipase C inhibitor D609 (50 microgram/ml). Pretreatment of D609 also effectively inhibited the translocation of protein kinase C betaI and betaII from the cytosol to the membrane and the activation of phospholipase D induced by Fas cross-linking, suggesting that 1, 2-diacylglycerol released from the cellular phosphatidylcholine pool through phosphatidylcholine-specific phospholipase C plays a major role in protein kinase C/phospholipase D activation. Anti-Fas monoclonal antibody failed to elicit phosphoinositide-specific phospholipase C activation and any changes in the intracellular Ca2+ level in A20 cells, indicating that the phosphoinositide-mediated pathway is not involved in this Fas signaling. Therefore, these results suggest that Fas-mediated phospholipase D activation may be a consequence of primary stimulation of phosphatidylcholine-specific phospholipase C and that phospholipase D may play a role in Fas cross-linking signaling downstream from phosphatidylcholine-specific phospholipase C.","is_dataset_classified":null,"base_score":2.833213344056216,"endowment":2.833213344056216,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"10395739","pmcid":null,"openalex_id":"https://openalex.org/W2066943142","authors":[],"funders":[{"funder_name":"Ministry of Education","grant_id":"219","title":null}],"total_grants":1,"fwci":1.1749,"citation_percentile":0.77650559,"influential_citations":0,"citation_trend":[],"oa_status":"closed","license":"https://www.elsevier.com/legal/tdmrep-license","oa_locations":[{"url":"https://api.elsevier.com/content/article/PII:S000398619991250X?httpAccept=text/xml","host_type":"publisher"},{"url":"https://api.elsevier.com/content/article/PII:S000398619991250X?httpAccept=text/plain","host_type":"publisher"},{"url":"https://doi.org/10.1006/abbi.1999.1250","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/10395739","host_type":"repository"}],"fields_of_study":["Protein Kinase Regulation and GTPase Signaling","Cell death mechanisms and regulation","Venomous Animal Envenomation and Studies","Animals","Bridged-Ring Compounds","Calcium","Cell Membrane","Cytosol","Dose-Response Relationship, Immunologic","Mice","Norbornanes","Phosphodiesterase Inhibitors","Phospholipase D","Phosphorylcholine","Receptors, Tumor Necrosis Factor","Thiocarbamates","Thiones","Time Factors","Tumor Cells, Cultured","Type C Phospholipases","fas Receptor"],"mesh_terms":["Animals","Bridged-Ring Compounds","Calcium","Cell Membrane","Cytosol","Dose-Response Relationship, Immunologic","Norbornanes","Phosphodiesterase Inhibitors","Type C Phospholipases","Phospholipase D","Phosphorylcholine","Thiocarbamates","Thiones","Time Factors","Tumor Cells, Cultured","Receptors, Tumor Necrosis Factor","fas Receptor","Mice"],"keywords":["Phospholipase C","Diacylglycerol kinase","Phosphoinositide phospholipase C","Phospholipase","Phospholipase D","Phosphatidylcholine","PLD2","Gq alpha subunit","Protein kinase C","Phospholipase A2","Biology","Molecular biology","Phosphatidylethanol","Biochemistry","Chemistry","Signal transduction","Phospholipid","Enzyme","G protein","Membrane"],"sdg_mappings":[],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-18T13:06:11.907022Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
