{"doi":"10.1002/oby.20444","title":"Association between obesity‐related gene <i>FTO</i> and ADHD","abstract":"<jats:sec><jats:title>Objective</jats:title><jats:p>Attention‐deficit/hyperactivity disorder (ADHD) is an etiologically complex heterogeneous behavioral disorder. Several studies have reported that ADHD subjects are more likely to be overweight/obese and that this comorbidity may be due to shared genetic factors. The objective of this study is to explore the association between ADHD and <jats:italic>FTO</jats:italic>, a gene strongly associated with obesity in genome‐wide studies.</jats:p></jats:sec><jats:sec><jats:title>Design and Methods</jats:title><jats:p>One tag SNP (single‐nucleotide polymorphism, <jats:italic>rs8050136</jats:italic>, risk allele A) in the <jats:italic>FTO</jats:italic> gene was selected and its association with ADHD was tested. Family‐based association tests (FBATs) were conducted with the categorical diagnosis of ADHD as well as behavioral and cognitive phenotypes related to ADHD. Furthermore, stratified FBAT analyses based on maternal smoking during pregnancy (MSDP) status were conducted.</jats:p></jats:sec><jats:sec><jats:title>Results</jats:title><jats:p>Statistically significant associations were observed between <jats:italic>rs8050136</jats:italic> and several of the traits tested in the total sample. These associations were stronger when the analysis was restricted to children who were not exposed to MSDP.</jats:p></jats:sec><jats:sec><jats:title>Conclusions</jats:title><jats:p>These exploratory results suggest the involvement of the <jats:italic>FTO</jats:italic> SNP <jats:italic>rs8050136</jats:italic> in modulating the risk for ADHD, particularly in those children who were not exposed to MSDP.
