However, most studies induce relatively high systemic inflammation, not directly compared with the more common low‐grade inflammatory events experienced in humans during the life course. Using mice, we investigated the effects of low‐grade systemic inflammation during an otherwise healthy early life, and how this may precondition the onset and severity of Alzheimer's disease (AD)‐like pathology. Our results indicate that low‐grade systemic inflammation induces sub‐threshold brain inflammation and promotes microglial proliferation driven by the CSF1R pathway, contrary to the effects caused by high systemic inflammation. In addition, repeated systemic challenges with low‐grade LPS induce disease‐associated microglia. Finally, using an inducible model of AD‐like pathology (Line 102 mice), we observed that preconditioning with repeated doses of low‐grade systemic inflammation, prior to APP induction, promotes a detrimental effect later in life, leading to an increase in Aβ accumulation and disease‐associated microglia. These results support the notion that episodic low‐grade systemic inflammation has the potential to influence the onset and severity of age‐related neurological disorders, such as AD.
","journal":"Glia","year":2024,"id":17178,"datarank":0.698479339602986,"base_score":3.2188758248682006,"endowment":3.2188758248682006,"self_citation_contribution":0.48283137373023016,"citation_network_contribution":0.21564796587275584,"self_endowment_contribution":0.48283137373023016,"citer_contribution":0.21564796587275584,"corpus_percentile":null,"corpus_rank":null,"citation_count":24,"citer_count":23,"citers_with_citation_signal":13,"citers_with_endowment":13,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":null,"is_data_producer":false,"deposit_databanks":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":123789,"name":"Imogen Targett","orcid":null,"position":1,"is_corresponding":false},{"id":123790,"name":"Adrian Olmos‐Alonso","orcid":null,"position":2,"is_corresponding":false},{"id":123791,"name":"Mariana Vargas‐Caballero","orcid":null,"position":3,"is_corresponding":false},{"id":123792,"name":"Diego Gomez‐Nicola","orcid":"0000-0002-5316-2682","position":4,"is_corresponding":false},{"id":123788,"name":"Monica Guerrero‐Carrasco","orcid":"0000-0002-5232-3608","position":0,"is_corresponding":false}],"reference_count":0,"raw_metadata":{"has_enrichment":true,"base_score":3.1780538303479458,"endowment":3.1780538303479458,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"38597386","pmcid":null,"openalex_id":"https://openalex.org/W4394692040","authors":[],"funders":[{"funder_name":"Medical Research Council","grant_id":"MR/P024572/1","title":"Is the microglial response in Alzheimer's disease determined by a dysfunctional balance of proliferation and survival?"},{"funder_name":"Gerald Kerkut Charitable Trust","grant_id":"","title":null}],"total_grants":2,"fwci":5.1198,"citation_percentile":0.95796489,"influential_citations":1,"citation_trend":[{"year":2024,"count":6},{"year":2025,"count":11},{"year":2026,"count":6}],"oa_status":"hybrid","license":"cc-by","oa_locations":[{"url":"https://onlinelibrary.wiley.com/doi/pdfdirect/10.1002/glia.24532","host_type":"journal"},{"url":"https://onlinelibrary.wiley.com/doi/pdfdirect/10.1002/glia.24532","host_type":"HYBRID"},{"url":"https://onlinelibrary.wiley.com/doi/pdfdirect/10.1002/glia.24532","host_type":"publisher"},{"url":"https://onlinelibrary.wiley.com/doi/pdf/10.1002/glia.24532","host_type":"publisher"},{"url":"https://doi.org/10.1002/glia.24532","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/38597386","host_type":"repository"},{"url":"https://doi.org/10.1101/2024.03.15.585224","host_type":""},{"url":"https://eprints.soton.ac.uk/488662/1/Guerrero-Carrasco_et_al_R2.pdf","host_type":""},{"url":"https://eprints.soton.ac.uk/488662/2/Glia_-_2024_-_Guerrero_Carrasco_-_Low_grade_systemic_inflammation_stimulates_microglial_turnover_and_accelerates_the_onset.pdf","host_type":""},{"url":"https://doi.org/https://doi.org/10.1101/2024.03.15.585224","host_type":""}],"fields_of_study":["Neuroinflammation and Neurodegeneration Mechanisms","Alzheimer's disease research and treatments","Immune Response and Inflammation","Medicine","Biology","Animals","Microglia","Alzheimer Disease","Inflammation","Mice, Transgenic","Lipopolysaccharides","Mice, Inbred C57BL","Mice","Disease Models, Animal","Receptors, Granulocyte-Macrophage Colony-Stimulating Factor","Male","Female","Amyloid beta-Protein Precursor","Brain","Amyloid beta-Peptides","Cytokines"],"mesh_terms":["Alzheimer Disease","Animals","Brain","Disease Models, Animal","Female","Inflammation","Lipopolysaccharides","Male","Mice, Inbred C57BL","Mice, Transgenic","Receptors, Granulocyte-Macrophage Colony-Stimulating Factor","Cytokines","Amyloid beta-Peptides","Amyloid beta-Protein Precursor","Microglia","Mice"],"keywords":["Microglia","Inflammation","Systemic inflammation","Neuroinflammation","Immunology","Medicine","Disease","Alzheimer's disease","Systemic disease","Pathology","Biology","Neuroscience","LPS","Proliferation","Alzheimers disease","Disease‐associated Microglia","Lipopolysaccharides","Male","Amyloid beta-Peptides","610","Brain","Mice, Transgenic","Mice, Inbred C57BL","Mice","Disease Models, Animal","Amyloid beta-Protein Precursor","Alzheimer Disease","Receptors, Granulocyte-Macrophage Colony-Stimulating Factor","616","Animals","Cytokines","Female"],"sdg_mappings":[{"sdg_number":3,"sdg_label":"3.
