{"doi":"10.1002/cam4.1228","title":"Phosphorylated <scp>ERK</scp> is a potential prognostic biomarker for Sorafenib response in hepatocellular carcinoma","abstract":"<jats:title>Abstract</jats:title><jats:p>Sorafenib, the only approved drug for hepatocellular carcinoma, acts as a remarkable inhibitor of Raf serine‐threonine kinases. However, Sorafenib is expensive, and clinical experience shows that it is not an effective treatment for many patients. Previous study has demonstrated that phosphorylated <jats:styled-content style=\"fixed-case\">ERK</jats:styled-content> (p<jats:styled-content style=\"fixed-case\">ERK</jats:styled-content>) is a key downstream component in the <jats:styled-content style=\"fixed-case\">RAF</jats:styled-content>/<jats:styled-content style=\"fixed-case\">MEK</jats:styled-content>/<jats:styled-content style=\"fixed-case\">ERK</jats:styled-content> signaling pathway. Here, we investigate whether p<jats:styled-content style=\"fixed-case\">ERK</jats:styled-content> is a useful biomarker for treating <jats:styled-content style=\"fixed-case\">HCC</jats:styled-content> with Sorafenib. In vitro cell viability assays showed that the efficacy of Sorafenib was distinctly different according to the level of p<jats:styled-content style=\"fixed-case\">ERK</jats:styled-content>. Furthermore, in established patient‐derived xenografts from <jats:styled-content style=\"fixed-case\">HCC</jats:styled-content> specimens, we found that the growth rate of tumors with high levels of p<jats:styled-content style=\"fixed-case\">ERK</jats:styled-content> was significantly decreased by Sorafenib treatment.
