We have studied this question in a large population of 119 EOAD patients (onset ⩽60 years) in which family history was carefully assessed and in 109 controls. We show that the APOE ϵ4 allele frequency is increased only in the subset of patients who belong to families where LOAD secondary cases are present. Our sampling scheme permits us to demonstrate that, for an individual, bearing at least one ϵ4 allele increases both the risk of AD before age 60 and the probability of belonging to a family with late‐onset affected subjects. Our results suggest that a subset of EOAD cases shares a common determinism with LOAD cases. © 1995 Wiley‐Liss, Inc.
","journal":"American Journal of Medical Genetics","year":1995,"id":599753,"datarank":0.49983067652628066,"base_score":3.332204510175204,"endowment":3.332204510175204,"self_citation_contribution":0.49983067652628066,"citation_network_contribution":0.0,"self_endowment_contribution":0.49983067652628066,"citer_contribution":0.0,"corpus_percentile":null,"corpus_rank":null,"citation_count":27,"citer_count":0,"citers_with_citation_signal":0,"citers_with_endowment":0,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":null,"is_data_producer":false,"deposit_databanks":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"fair_score":null,"fair_percentile":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":62326,"name":"Dominique Campion","orcid":"0000-0003-1916-7890","position":1,"is_corresponding":false},{"id":1537182,"name":"Maria Martinez","orcid":null,"position":2,"is_corresponding":false},{"id":17405,"name":"Alexis Brice","orcid":"0000-0002-0941-3990","position":3,"is_corresponding":false},{"id":1537183,"name":"Sandrine Tardieu","orcid":null,"position":4,"is_corresponding":false},{"id":618059,"name":"Didier Hannequin","orcid":"0000-0003-2662-0999","position":5,"is_corresponding":false},{"id":17554,"name":"Yves Agid","orcid":null,"position":6,"is_corresponding":false},{"id":168463,"name":"André Delacourte","orcid":null,"position":7,"is_corresponding":false},{"id":54821,"name":"Françoise Clerget‐Darpoux","orcid":"0000-0003-4816-6619","position":8,"is_corresponding":false},{"id":34703,"name":"Marie‐Christine Chartier‐Harlin","orcid":"0000-0001-6416-6526","position":9,"is_corresponding":false},{"id":241322,"name":"Jordi Pérez‐Tur","orcid":"0000-0002-9111-1712","position":0,"is_corresponding":false}],"reference_count":0,"raw_metadata":{"has_enrichment":true,"resolved":true,"title":"Evidence for apolipoprotein E ϵ4 association in early‐onset Alzheimer's patients with late‐onset relatives","abstract":"
Abstract
Recently several reports have extended the apolipoprotein E (APOE) ϵ4 association found in late‐onset Alzheimer's disease (LOAD) patients to early‐onset (EO) AD patients.
