Human epidermal growth factor receptor 2 overexpression, found in 20–30% of breast cancer patients is associated with aggressive tumour behavior, and poorer prognosis. Trastuzumab (Herceptin), a biologically engineered, humanized immunoglobulin-1, anti-HER2 monoclonal antibody (mAb) developed by Genentech/ Roche, directed against the extracellular domain of HER2. It is the first commercially available mAb-based therapy for the treatment of breast cancer. Trastuzumab has been extensively trialed and now is used in the early and metastatic settings. Ado-trastuzumab emastasine (T-DM1, Genentech Roche) is a novel antibody–drug conjugate, whereby trastuzumab (T) is linked to mertansine (DM1). It is currently undergoing multicentered phase 3 trials exploring its full role within the treatment of Her2 overexpression breast cancer, with promising early results.","is_dataset_classified":null,"base_score":1.9459101490553132,"endowment":1.9459101490553132,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"26207759","pmcid":null,"openalex_id":"https://openalex.org/W1594988246","authors":[],"funders":[],"total_grants":0,"fwci":null,"citation_percentile":null,"influential_citations":0,"citation_trend":[{"year":2016,"count":1},{"year":2018,"count":1},{"year":2023,"count":1},{"year":2025,"count":2},{"year":2026,"count":1}],"oa_status":"closed","license":null,"oa_locations":[{"url":"https://onlinelibrary.wiley.com/doi/pdf/10.1002/9783527682423.ch72","host_type":"publisher"},{"url":"https://doi.org/10.1002/9783527682423.ch72","host_type":""}],"fields_of_study":["HER2/EGFR in Cancer Research","Monoclonal and Polyclonal Antibodies Research","Peptidase Inhibition and Analysis"],"mesh_terms":[],"keywords":["Trastuzumab","Trastuzumab emtansine","Medicine","Breast cancer","Antibody-drug conjugate","Humanized antibody","Monoclonal antibody","Oncology","Metastatic breast cancer","Cancer","Internal medicine","Antibody","Cancer research","Immunology"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"No poverty"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-18T12:43:13.520769Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
