This chapter outlines the basic principles of fluorescence and fluorescent sensing. It explains common classes of fluorophores and the mechanisms by which fluorescent sensors operate. There are many characteristics of fluorescence that are advantageous for application in imaging and spectroscopy. Fluorescence imaging is amongst the most popular and widely used techniques for molecular imaging. Fluorescence microscopy has become more powerful due to developments such as fluorescence lifetime imaging microscopy and super resolution microscopy. By way of generalisation, a small-molecule fluorescent sensor can be considered to consist of two parts: the fluorescence-generating group, or fluorophore; and the sensing moiety that interacts with the analyte of interest.
