Early biopharmaceutics assessment is generally based on minimal testing to provide a fit for purpose data set. More detailed pharmacological and physicochemical profiling is undertaken later in drug discovery as compounds are optimised towards candidate drug selection. Pharmacological activity in response to the extent and duration of target engagement is monitored in enzyme, cell or animal models to establish dose-response relationships and define exposure profile requirements. Biopharmaceutics assessment of potential drug candidates is underpinned by understanding of fundamental properties and their influence on exposure: solubility, permeability, lipophilicity, pKa, molecular size and solid form. The biopharmaceutics classification system uses the two factors that govern the rate and extent of drug absorption: solubility and intestinal permeability.
