Photobleaching is the photo‐induced change in a fluorphore that abolishes that molecule's fluorescence. The different characteristics of green fluorescent protein (GFP) chimeras in a cell can be studied by FRAP, in which a selected region of the cell is photobleached with intense light. The movement of unbleached molecules into a photobleached region is quantified by imaging with an attenuated light source. The movement of molecules between cellular compartments can be determined by FLIP, in which the same region of a cell expressing a GFP chimera is repeatedly photobleached. The loss of fluorescence from regions outside the photobleached region is monitored to characterize the movement of a protein. Together these two techniques are providing fundamentally new insights into the kinetic properties of proteins in cells.
","journal":"Current Protocols in Cell Biology","year":2003,"id":29968,"datarank":5.680672919167178,"base_score":4.7535901911063645,"endowment":4.7535901911063645,"self_citation_contribution":0.7130385286659547,"citation_network_contribution":4.967634390501224,"self_endowment_contribution":0.7130385286659547,"citer_contribution":4.967634390501224,"corpus_percentile":null,"corpus_rank":null,"citation_count":115,"citer_count":92,"citers_with_citation_signal":86,"citers_with_endowment":86,"datacite_reuse_total":0,"is_dataset":false,"is_dataset_confidence":null,"is_oa":false,"file_count":0,"downloads":0,"has_version_chain":false,"published_date":null,"algorithm_id":"datarank_citation_only_1hop_v6","ranking_scope":"data_only","authors":[{"id":163950,"name":"Nihal Altan","orcid":null,"position":1,"is_corresponding":false},{"id":62806,"name":"Jennifer Lippincott‐Schwartz","orcid":"0000-0002-8601-3501","position":2,"is_corresponding":false},{"id":163948,"name":"Erik L.
