Whereas treatment with the nucleoside purine analogues cladribine and pentostatin results in lengthy remissions in nearly all patients with HCL, most patients will experience relapse while a small percentage of patients' disease fails to respond to therapy in the first place. Retreatment with a purine nucleoside analogue often leads to an effective but limited response. For decades, few other viable therapeutic options were available to these patients who required retreatment. Recently, new insights into the mechanism of disease of HCL have led to research in new potential treatment agents, either alone or with a purine nucleoside analogue. Clinical trials with rituximab, bendamustine, and conjugate immunotoxins will reveal what role these therapies will have in HCL treatment. A better understanding of the BRAF/MEK/ERK pathway and the B-cell signaling pathway has allowed further exploration into the novel drugs vemurafenib, dabrafenib, trametinib, and ibrutinib.","is_dataset_classified":null,"base_score":3.4339872044851463,"endowment":3.4339872044851463,"datacite_reuse_total":0,"file_count":0,"downloads":0,"views":0,"has_version_chain":false,"is_dataset":false,"is_oa":false,"pmid":"26513168","pmcid":null,"openalex_id":"https://openalex.org/W1891896135","authors":[],"funders":[],"total_grants":0,"fwci":1.4544,"citation_percentile":0.81114297,"influential_citations":0,"citation_trend":[{"year":2016,"count":1},{"year":2017,"count":5},{"year":2018,"count":3},{"year":2019,"count":4},{"year":2020,"count":3},{"year":2021,"count":3},{"year":2022,"count":6},{"year":2023,"count":3},{"year":2024,"count":1},{"year":2025,"count":1}],"oa_status":"closed","license":null,"oa_locations":[{"url":"https://oncology.jamanetwork.com/ama/content_public/journal/oncology/934824/crv150011.pdf","host_type":"publisher"},{"url":"https://doi.org/10.1001/jamaoncol.2015.4134","host_type":"journal"},{"url":"https://pubmed.ncbi.nlm.nih.gov/26513168","host_type":"repository"}],"fields_of_study":["Chronic Lymphocytic Leukemia Research","Toxin Mechanisms and Immunotoxins","PI3K/AKT/mTOR signaling in cancer","Antineoplastic Agents","Biomarkers, Tumor","Diffusion of Innovation","Drug Resistance","History, 20th Century","History, 21st Century","Humans","Leukemia, Hairy Cell","Molecular Targeted Therapy","Patient Selection","Recurrence","Signal Transduction","Treatment Outcome"],"mesh_terms":["Antineoplastic Agents","Diffusion of Innovation","Drug Resistance","Humans","Leukemia, Hairy Cell","Recurrence","Biomarkers, Tumor","Signal Transduction","Treatment Outcome","Patient Selection","History, 20th Century","History, 21st Century","Molecular Targeted Therapy"],"keywords":["Cladribine","Medicine","Pentostatin","Hairy cell leukemia","Purine analogue","Vemurafenib","Nucleoside analogue","Ibrutinib","Bendamustine","Rituximab","Trametinib","Oncology","Leukemia","Dabrafenib","Pharmacology","Chronic lymphocytic leukemia","Internal medicine","Cancer research","Nucleoside","Purine","Lymphoma","MAPK/ERK pathway","Melanoma","Kinase"],"sdg_mappings":[{"sdg_number":0,"sdg_label":"Good health and well-being"}],"linked_datasets":[],"clinical_trials":[],"software_tools":[],"database_accessions":[],"source":"live","citation_network_status":"fetched"},"created_at":"2026-08-06T17:46:23.171262Z","pmid":null,"pmcid":null,"fwci":null,"citation_percentile":null,"influential_citations":0,"oa_status":null,"license":null,"views":0,"total_file_size_bytes":0,"version_count":0,"fair_f":null,"fair_a":null,"fair_i":null,"fair_r":null,"fair_zscore":null,"fair_rationale":null,"fair_model":null,"fair_agent_version":null,"fair_fulltext_source":null,"fair_has_llm":null,"fair_computed_at":null,"clinical_trials":[],"software_tools":[],"db_accessions":[],"linked_datasets":[],"topics":[]}
